Spontaneously Pancreatic Changes in Diabetic WBN/Kob Rats

Abstract
WBN/Kob rat is an animal model for human diabetes. WBN/Kob rats have significant characteristics compared with other diabetic animal models. Not only endocrine portion but also exocrine portion of pancreas of WBN/Kob rate were destroyed by distinct hemorrhage and fibrosis. However the etiology of pancreatic destruction leading to diabetes is not cleary understood. Also We observed pancreas of WBN/Kob rats histologlcally and immunohietochemically to elucidate the relationships between onset of diabetes and pancreatic destruction, especially in pancreatic endocrine cells. WBN/Kob rate were used from the age of 1-24 months. High blood glucose and high level of plasma triglyceride and cholesterol were distinct in diabetic WBN/kob rats. Blood glucose level of WBN/Kob rats were slightly high. The pancreas of male WBN/Kob rate, over the age of 8 months, were showed hemorrhage, deposition of hemosiderin, infiltration of inflammatory cells around pancreatic ducts and blood vessels. The number and size of pancreatic islets were reduced according to the proliferation of fibrous tissue. In female WBN/Kob rats showed hemorrhage, deposition of hemoeiderin and fibrosis from 18 months but these findings were very slightly and not all female WBN/Kob rate. But no female WBN/Rob rats showed diabetic symptoms. The relative populations of both pancreatic A and B cells in male diabetic WBN/Kob rats decreased after onset of diabetes. We conclude the onset of diabetes of WBN/Kob rate come from the destruction of pancreatic A and B cells by wide spread fibrosis and inflammatory reactions. Also WBN/Kob rats were considered as an useful animal models for studying diabetic complications without insulin therapeutics.
